New photo-CIDNP hyperpolarization methods for NMR of studies of small molecules and biomolecules at ICSN and Eurofins.

NMR spectroscopy is an extremely powerful analytical method for characterising molecules of various origins (synthetic, biological) and their mixtures in terms of structure, dynamics and molecular interactions, but also for determining the absolute or relative concentrations of the elements composing a mixture and the presence or absence of a particular compound. This spectroscopy has therefore become a major technique in both academic research and industry in health, agri-food, energy, etc. However, this technique suffers from a fundamental limitation in terms of sensitivity, severely limiting its ability to study compounds at low concentrations. The aim of the thesis will be to exploit and improve an original hyperpolarisation approach based on the photo-CIDNP technique. The photo-CIDNP technique [1] consists of illuminating a sample containing a dye with suitable photophysical properties, causing it to enter an excited triplet state, a great source of polarization. The molecular interactions between the dye and the molecule to be hyperpolarized leads to polarization transfer to the molecule of interest, thereby boosting its NMR signal. During the thesis, we will aim to develop several methods to increase the NMR signals of a larger number of spins/atoms. We will adapt these methods to improve the detection of small molecules or peptides in complex mixtures or to better characterize the surface of proteins in relation to chemical and post-translational modifications, related to pharmaceutical and therapeutical applications.
The PhD student will be trained to state-of-the-art NMR spectroscopy and liquid chromatography applied to small molecules and proteins and will gain expertise in photophysics and in situ illumination central to photochemistry and photobiology. S.he will have privileged access to exceptional NMR equipments at ICSN (ten spectrometers up to 950 MHz) and various analytical techniques at Eurofins. The PhD will be funded by an industrial collaboration with Eurofins located in Les Ulis, 20 mn by car from ICSN lab in Gif-sur-Yvette. The PhD student will share his.her time between ICSN (60%) and Eurofins (40%).

- Profil : The candidate should ideally have a background in physical chemistry and analytical chemistry with strong interests in developing new analytical techniques for small molecules and biomolecules in line with pharmaceutical and therapeutical applications. The candidate will also express deep interest in working in an academic/industrial collaborative environment.
