Characterizing the fibrillation of amyloid proteins, both in vitro and in cellulo

This project aims at characterizing the aggregation process of two amyloid proteins (IAPP, involved in type 2 diabetes, and PSMα3, secreted by the pathogen S. aureus) under conditions of increasing complexity: (i) in vitro in simple buffers, and (ii) in cellulo in the presence of cells. Importantly, the later will allow to reflect physiological conditions and to apprehend the role of cell membranes in eventually modulating the fibrillation process. Fluorescence spectroscopy will be used to study the kinetics of fibrillation, with a critical work on optimizing the experimental conditions (type of fluorophore, concentration, temperature, etc). Then, the aggregates and fibers formed at given times, in the presence or absence of cells, will be characterized morphologically by microscopies (e.g. electron microscopy (TEM), atomic force microscopy (AFM) and/or fluorescence microscopy). Finally, fibrillation will be correlated with the cytotoxicity of these amyloid proteins, using cell viability assays to associate the formation of specific entities with this deleterious biological activity.
